The Opsonophagocytic Assay Kit (FX2002) measures the level (titer) of opsonic antibody activity in human sera. This kit utilizes polymorphonuclear (PMN) leukocytes (see notes below), 3-4 week baby rabbit complement, test antibody, and an antigen target. The antigen for this kit is native whole cell fluorescently labeled S. pneumoniae. 13 serotypes are provided (PREVNAR13®).
The interaction of antibody with antigen forms an antigen/antibody complex and leads to the activation of the complement cascade. This activation of the complement converts C1 to C3b, a protein that serves as an opsonin when bound to an antigen surface. Opsonization mediates phagocytosis, and the antigen (fluorescent bacteria) will be consumed and internalized by the phagocyte (PMN). The cells are then analyzed using a Flow Cytometer, and fluorescent signal is measured. This fluorescent signal is used to calculate a titer.
